Genes overexpression targets for enhancement of heterologous production of diverse terpenoids in Escherichia coli |

Objective and Background Terpenoids (also known as isoprenoids) belong to a large class of natural metabolites mainly produced in plants and derived from five carbon isoprene units (Withers and Keasling, 2007). They are naturally produced by two different metabolic pathways: 1) The MEP (methyl erythritol phosphate) pathway started by a combination of one pyruvate molecule by two glyceraldehyde 3-phosphate molecules; and 2) The MVA (mevalonate) pathway started from acetyl-CoA and acetoacetyl-CoA molecules. Terpenoids are widely used in the industry and pharmacy and inspire researchers to develop their microbial production (Vickers et al., 2014). Recently, as a consequence of advancements in science and technology, genome-scale metabolic models (GEMs) enable researchers to easily understand the genotype-phenotype relationship in microbial metabolisms and identify potential modifications for improvement of desired metabolite production. For this purpose, several in silico algorithms have been developed so far. Most of these algorithms, like “OptKnock” (Burgard et al., 2003), are based on gene deletion but just a few of them introduce the candidates for gene overexpression (Choi et al., 2010; Jian et al., 2016; Park et al., 2012). In this research, employing the systems biology tools a new bottom-up approach for enhancement of terpenoids microbial production in Escherichia coli is developed. Firstly, the method by which the overexpression reaction candidates are identified is presented, and then the candidates to enhance different terpenoids biosynthesis are compared. At the end, the supreme 17 common reactions and their associated gene targets for overexpression are reported. |
