توصیف مجدد و معتبر شناختن گونهPlectonchus zondagi (Dale, ۱۹۶۷) Abolafia, Alizadeh & Khakvar, ۲۰۱۶ (Rhabditida, Brevibuccidae) از ایران |

بر اساس جمعیت یافت شده از Plectonchus zondagi در شمال ایران، این گونه توصیف مجدد شده است. مشخصات ریخت‌شناسی جدید بر اساس مشاهدات میکروسکوپ نوری و میکروسکوپ الکترونی نگاره (SEM) و آنالیزهای مولکولی بر اساس ناحیه تقریباً کامل زیر واحد کوچک (SSU) و ناحیه D2/D3 زیر واحد بزرگ (LSU) DNA ریبوزومی ارائه شده است. این گونه با داشتن خصوصیاتی از جمله لبهای مجاور در هم ادغام شده، آمفیدهای پسین، استوما با چیلو-ژیمنوستوم بزرگ، متاکورپوس در مری به ندرت متورم، دستگاه تناسلی ماده دارای یک تخمدان رو به جلو و فاقد کیسه عقبی رحم، نر با اسپیکولهای زاویهدار، گوبرناکولوم سیگموئیدی، دم در هر دو جنسیت مخروطی با انتهای تیز و فشرده و دارای یک حلقه کوتیکولی برگشته بین قسمتهای متصل و جدا شده کوتیکول، متمایز میشود. تجزیه و تحلیل‌های تبارشناسی مولکولی بر اساس توالی‌های زیر واحد کوچک (1409 جفت باز) و ناحیه D2/D3 زیر واحد بزرگ (969 جفت باز) ریبوزومی نشان داد که جنس Plectonchus با دو جنس Brevibucca و Cuticonema خویشاوندی نزدیکی دارد. همچنین، P. zondagi با اعضای خانواده Brevibuccidae در یک کلاد با حمایت بالا قرار گرفت که با توجه به شباهت در موقعیت و نوع سیستم تولید مثل ماده و شباهت در جدا شدگی کوتیکول، این یافته با شاخصهای ریخت‌شناسی نیز حمایت میشود. به طور کلی، نتایج این پژوهش نشان دهنده این است که جایگاه تاکسونومی مناسب این گونه در جنس Plectonchus تحت خانواده Brevibuccidae میباشد. |

Plectonchus zondagi is redescribed based on the population found in the north of Iran. New morphological characterizations based on light and scanning electron microscopy and molecular analyses based on near full-length of small subunit (SSU) and D2/D3 segment of large subunit (LSU) ribosomal DNA are provided. This species is characterized by having a lip region with lips fused in pairs, amphids posterior, stoma with large cheilo-gymnostom, pharynx with scarcely swollen metacorpus, female reproductive system prodelphic, absence of post vulval uterine sac, male with angular spicules and sigmoid gubernaculum and tail in both sexes conoid with acute tip bearing a refringent cuticular ring between the attached and detached parts of the tail cuticle. Phylogenetic analyses based on the sequences of small subunit (1409 bp) and D2/D3 segment of large subunit (969 bp) ribosomal RNA genes showed that the genus Plectonchus have a relationship with Brevibucca and Cuticonema. Thus, P. zondagi forms a well-supported clade with members of the family Brevibuccidae that is supported morphologically by having similarity in the position and type of the reproductive system of females, and detached cuticle. P. zondagi was synonymized with P. molgos in 2016 by Abulafia et al., but in this study, these two species are differentiated by the acute tail end in both males and females of the recovered population, and the rounded tail tip in P. molgos. |
